1029   RUMEN BACTERIA MEDIUM

Mineral solution (see below)  38.0     ml
K2HPO4  0.3     g
Trypticase peptone (BD-BBL)  2.0     g
Yeast extract  0.5     g
VFA solution (see Medium No. 133)  3.1     ml
Hemin  1.0     mg
Glucose  0.5     g
Maltose  0.5     g
Cellobiose  0.5     g
Starch, soluble  0.5     g
Glycerol  0.5     g
Na2CO3  4.0     g
Resazurin  1.0     mg
L-Cysteine·HCl·H2O  0.25    g
Na2S·9H2O  0.25    g
Distilled water  960.0     ml
Adjust pH to 6.7 - 6.8; gas atmosphere: 100% CO2.
Dissolve ingredients except carbohydrates, Na2CO3, cysteine and sodium sulfide in distilled water, boil for 5 min, then cool to room temperature while gassing with 100% CO2. Add Na2CO3 and equilibrate the medium with the gas to pH 6.8. Distribute under CO2 in rubber-stoppered tubes (i.e. Hungate tubes) and autoclave. Thereafter add carbohydrate mixture, cysteine and sodium sulfide from concentrated sterile stock solutions (prepared anaerobically under N2 gas and autoclaved).
