Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.

1286 HALANAEROARCHAEA MEDIUM WITH YEAST EXTRACT AND GLUCOSE

Base salt medium (see Medium No. 1207) 1.0 L

Autoclave. After cooling, add the following solutions (autoclaved or *filter-sterilized):

1 M MgSO₄ solution	`1.0`	ml
Trace element solution (see Medium No. 1079)	`1.0`	ml
Selenite-tungstate solution* (see Medium No. 431)	`1.0`	ml
Trace vitamins* (see Medium No. 197)	`5.0`	ml
8% NaHCO₃ solution*	`50.0`	ml

To complete the medium, aseptically and anaerobically distribute the medium into culture vessels containing steam-sterilized sulfur (see below, approximately 5.0 g/L) under an argon gas stream, and seal with butyl rubber stoppers. Then, add per liter the following solutions (autoclaved or *filter-sterilized):

10% Yeast extract solution	`1.0`	ml
1.0 M Glucose solution*	`10.0`	ml
5% Na₂S·9H₂O solution	`2.5`	ml

Sterilizationofsulfur:
Steam sulfur for 3 hr, or autoclave at 105C for 1 hr, on each of 3 successive days.
